Monoclonal antibodies (mAbs) are the most accelerating drug products on the market. With an increasing demand of mAbs, a significant burden has been placed on the mAbs capture and downstream process. The Protein A column chromatography process, which can reach over 90% purity, is the overall golden standard process for the purification of mAbs. However, before the Protein A column capture step, the cells producing the mAbs needs to be removed by filtration or centrifugation in order to prevent column clogging issues. This step will decrease the overall yield, is costly, can take many hours, and also to increase host cell protein (HCP) levels and proteolytic activity in the clarified material, due to cell lysis demanding following challenging purification steps.

Our MAGic methodology is a one-step batch separation product-recovery process, directly applied on the cell broth, and provides increased product yield and savings in terms of time, material, and other resources, while also reducing the HCP levels. The methodology includes a magnetized resin, our MAGicBeads dedicated for mAb capture, together with specially designed magnetic separators, our MAGicSep units, optimized for efficient separation of the magnetized resin. Our products gives unique scalability for the lab, pilot and the process scale.

  • MAGicBeads mAb

    MAGicBeads mAb is a magnetic bead coupled with an alkali resistant protein A ligand optimized for purification of mAbs directly from the cell broth. The high bindning capacity, stability of the matrix and alkali resistance of the ligand makes the resin suitable for repeated purification cycles without loosing performance when cleaning with 0.1-0.2M NaOH between the runs.

  • MAGicBeads PrtA

    MAGicBeads PrtA is an agarose-based magnetic bead coupled with an recombinant protein A ligand for capture and recovery of monoclonal and polyclonal antibodies directly from complex feedstocks.

  • MAGicBeads SerA

    MAGicBeads SerA are optimized for the purification of polyclonal antibodies from serum and monoclonal antibodies from ascites fluid samples. The magnetic beads have a binding capacity of 60 mg human or rabbit IgG per ml settled beads.

  • MAGicSep Lab, Pilot and Process

    The MAGicSep Lab and Pilot units are designed for durability with a lacquered plastic shell and nickel plated neodymium magnets placed safely inside. They fit standard test tubes and are available in sizes from 1 ml to liters. The MAGicSep Process units are made for efficient capture of 1-40 liters of MAGicBeads from thousands of litres of complex feedstocks.

CELLS

Today several projects within therapy are based on using viable cells as personalized medicines and these cells have to be pure and active. The separation and sorting processes of the cells is thus of critical nature.

Antibodies-image

Numerous cell isolation and sorting techniques have been developed for benchtop and clinical settings that are based on either physical properties of the cell or on cell-surface affinity. Standard separation techniques based on the cells physiological properties include filtration, centrifugation and sedimentation. However, in the field of cell therapy the cell size or density differences are not significant and effective cell separation is hindered. Therefore other methods are used.

Isolation techniques that are based on the cell-surfaces antigen binding such as fluorescence activated cell sorting (FACS) and magnetic activated cell sorting (MACS) is now popular and attractive. Because of its specificity, flexibility, ease-of-use and scalability MACS is currently the technique with the greatest potential for cell separation in the cell therapy field. Until now, effective magnetic-based cell separation methodologies are present for up to 100ml sample volumes.

Our novel MAGic methodology including magnetized resins and magnetic separators for all scales of purification of cells for therapy use. The technique offers a unique scalability from the lab bench through pilot and to the final process scale.

    Surface activated magnetic beads for covalent couplig of antibodies. By coupling cell-surface antigen active antibodies, the magnetic beads can be used both in positive selection and in negative selection (untouched) approaches of cell separation. We offer customized covalent coupling service of antibodies to the activated beads.

Magnetic resins

MAGic bioprocessing provides magnetic agarose-based resins that are used to recover biologics or other biomolecules directly from crude sample mixtures eliminating the need of centrifugation or filtration. Our MAGicBeads are highly porous which provides a large available surface area (50m2/ml) resulting in exceptional high binding capacities, similar to column resins. The beads are also strongly magnetized (40emu/g) and stable due to our proprietary manufacturing method which provides consistent and fast magnetic separation even in large sample volumes (litres).

MAGic bioprocessing provides magnetic separators (MAGicSep) that are optimized for easy and efficient capture of the magnetic resin from crude sample mixtures. The standard units are designed for durability with a lacquered plastic shell and nickel plated neodymium magnets placed safely inside. They fit standard test tubes and are available in sizes from 1 ml to liters. The larger custom units are proprietary designed magnetic filters (MAGicSep Process) made for efficient capture of 1-40 liters of magnetic beads from infinite volumes of complex feedstocks.

Super-paramagnetic porous beads coupled with an alkali resistant protein A ligand. Optimized for purification of monoclonal antibodies directly from cell culture harvests, thus eliminating centrifugation or filtration, such as from hybridoma or CHO cells, where sanitization of beads with NaOH is desired between the uses. High bindning capacity and fast magnetic response makes the beads suitable for separation even in large-volume samples.

ACTIVATED BEADS

Surface activated magnetic agarose beads employing our patented coupling chemistry. Any ligand containing amino groups or thiols, e.g. proteins and peptides can be covalently bound to the beads using a simple process. All steps are performed at physiological conditions and room temperature, without using any hazardous reagents or organic solvents. After coupling of the ligand, the magnetic beads can be used for affinity purification and immunoprecipitation.

MAGicSEP

DURABLE MAGNETIC SEPARATORS FOR LAB, PILOT AND PROCESS SCALE

The MAGicSep series of laboratory magnetic separators have been optimized for use with MAGicBeads and are ideal for most bioseparation applications. The LAB and PILOT units are designed for durability with a lacquered plastic shell and nickel plated neodymium magnets placed safely inside. The separators fit standard test tubes and are available in sizes from 1 ml to 500 ml. The combination of MAGicBeads and MAGicSep is a simple biomagnetic separation platform that requires no special training to use. The separators require very little maintenance and several batches can be processed in parallel using the same unit, making the running costs very low. The larger custom units are proprietary designed magnetic separators (MAGicSep Process) made for efficient capture of 1-40 liters of magnetic beads from infinite volumes of complex feedstocks.

MAGicLAB

At the research lab, small-scale processing of peptides, proteins, antibodies or cells from crude sample mixtures like serum, whole blood, ascites, food-samples or unclarified cell cultures can be performed in volumes from 100 µl to 2 ml. By using 10-200 µl of MAGicBeads together with the MAGicSep LAB cube or LAB rack enables fast, reliable and easy separation of up to 6 mg of proteins per each tube via serial or parallel-process manual configuration.

Our MAGicBeads are also compatible with robotic system such as the KingFisher™ Flex unit (Thermo Fisher) allowing the use of our beads in automatic setups for high-throughput screening.

MAGicPILOT

During the development phase, medium-scale processing of peptides, proteins or antibodies from clarified- or unclarified cell-culture mixtures is performed in sample volumes from 5 ml up to a few liters. The use of the MAGicSep PILOT 50 or MAGicSep PILOT 500 together with 200µl – 30ml of settled MAGicBeads offers a durable, convenient and fast separation platform of up to gram quantities of proteins per each bottle via serial or parallel-process manual configuration.

By combining the different MAGicSep units, low expressed target peptides and proteins can be captured from a large volume and then washed and eluted in a smaller sized vessel, thus achieving a significant product concentration.

MAGicPROCESS

During the production phase of biopharmaceuticals, large-scale recovery of the product from an unclarified cell-culture is performed in a one step clarification and purification by using the propietary designed MAGicSep PROCESS units together with the affinity-coupled MAGicBeads. The magnetic filter separators are capable to capture up to 40 liters of MAGicBeads at high flow rates making them suitable to process up to kilo grams of proteins or antibodies per purification cycle.

By utilizing the magnetic-based capture approach, the biopharmaceutical can be recovered directly from the cell-containing culture with the same or better quality as for a conventional 2-step clarification and purification mode using depth filters and column chromatography.
