Human renal cell line ciPTEC – Radboud University

ciPTEC (conditionally immortalized Renal Proximal Tubule Epithelial Cell) is a novel cell line developed by Radboud University. ciPTEC is conditionally immortalized by using SV40 large T antigen and human telomerase reverse transcriptase (hTERT). ciPTEC differs from existing cell lines because it stably expresses most transporters.

ABCC2 = multidrug resistance protein 2 (MRP2)
ABCC4 = multidrug resistance protein 4 (MRP4)
ABCG2 = breast cancer resistance protein (BCRP)
SLC22A2 = organic cation transporters (OCT2)
SLC22A6 = organic anion transporters (OAT1)
SLC22A8 = organic anion transporters (OAT3)
SLC47A1 = multidrug and toxin extrusion transporter (MATE1)
SLC47A2 = multidrug and toxin extrusion transporter (MATE2-k)
SLCO4C1 = organic anion transporting polypeptide (OATP-H)

Reliable prediction of drug-induced kidney injury (DIKI) is a major issue during drug development. 1 out of 3 lead compounds lead to organ toxicity and 1 out of 5 compounds lead to renal toxicity during phase 3 of drug development. ciPTEC expresses all relevant drug transporters. This cell line provides a robust and predictive model for ADME/tox studies during drug development.
ciPTEC has the potential to reduce animal experiments in the future as highly predictive in vitro renal toxicity model
